چکیده

The specificity of cathepsin G, a serine neutral proteinase from human neutrophil leucotyes, was determine dby its action on the insulin B chain. The most susceptible bonds were Phe-24-Phe-25, Leu-15-Tyr-16 and Tyr-16-Leu-17. Other bonds hydrolysed were Leu-6-Cys(O3H)-7, Leu-11-Val-12, Leu-17-Val-18 and Phe-25-Tyr-26. These results suggest that the specificity of cathespin G is closer to that of pig chymotrypsin C than ox Chymotrypsin A. Tables listing amino acid composition, N-terminal residue, and yields of isolated peptides have been deposited as Supplementary Publication SUP 50 075 (8 pages) at the British Library Lending Division, Boston Spa, Wetherby, West Yorkshire LS23 7B2, U.K., from whom copies can be obtained on the terms indicated in Biochem. J. (1977) 161,1.

Effects of Endurance Training on the Expression of Cathepsin B (CTSB) and Cathepsin L (CTSL) genes in the Adipose Tissue of Mice with a High-Fat Diet

Introduction: In high-fat diet-induced obesity, the levels of cathepsin L (CTSL) and cathepsin B (CTSB) increase in adipocytes, resulting in insulin resistance in the adipose tissue. In this study, the preventive effect of endurance training on the gene expression of CTSL and CTSB was investigated in the adipose tissue of mice with a high-fat diet. Materials and Methods: Twenty-one male mice (a...

Proteolytic activity and cleavage specificity of cathepsin E at the physiological pH as examined towards the B chain of oxidized insulin.

Proteolytic activity and cleavage specificity of cathepsin E were investigated in a wide range of pHs from 3.0 to 10.5 using the B chain of oxidized insulin as substrate. Contrary to the previous notion that cathepsin E is virtually inactive above pH 6, significant proteolytic activity was observed at pH 7.4 and above. Further, cleavage specificity appeared to change significantly with pH and r...

The specificity of bovine spleen cathepsin S. A comparison with rat liver cathepsins L and B.

The peptide-bond-specificity of bovine spleen cathepsin S in the cleavage of the oxidized insulin B-chain and peptide methylcoumarylamide substrates was investigated and the results are compared with those obtained with rat liver cathepsins L and B. Major cleavage sites in the oxidized insulin B-chain generated by cathepsin S are the bonds Glu13-Ala14, Leu17-Val18 and Phe23-Tyr26; minor cleavag...

Proteases Detection of invitro Culture of Midgut Cells from Hyalomma anatolicum anatolicum (Acari: Ixodidae)

Proteases play a key role in protein digestion in ticks and other haematophagous insects. Our understanding of blood meal digestion in digestive system of ticks can be very useful for better understanding of basic rules for control of ticks. Cells of the midgut endocytose blood components. Blood proteins uptake by midgut cells, suggesting the presence of proteases in the midgut cells. In this...
